I'm in Tier 2 General Visa since June 2013.
I want to change my designation now. Is that possible? Also I'm willing to change the sponsor.
Please suggest me is that possible to change different designation, different sponsor?

Sure. Must meet requirements of whatever SOC code. New employer would need to conduct RLMT. You are not 'tied' to your current employer in the same way as ICT. Why would you think that?

You would need to apply from within the UK to avoid the 12 month cooling off period and you would need an unrestricted CoS.

It means if you leave your job in UK and go back to your home country for whatever reason
and try to re-apply for Tier 2(G) from home country, you need 12
months cooling off period (you can't apply before 12 months).

To avoid this, if you are applying inside UK before your visa expired, you need unrestricted
CoS instead restricted CoS that you got when you applied from outside of UK.
