i am applying in the process of citizenship. Does the refree sign the photos?

No they don't. To apply for citizenship, both of your referees have to sign in the referees section on the AN form.

It is only when applying for a british passport (to which you can apply after your citizenship application has been successful), that your referee has to sign on the photo.
