Professional referee

Hi,

We are going to apply for naturalisation based on Tier 2 + 1 year of ILR route.
I am a university lecturer. May I use a colleague of mine, a professor of our university as my ‘professional referee’? He is not my head of department, or line manager, just a colleague.

Can he be used as a professional referee for my wife and our 21y.o. son. He knows them as friend rather than a teacher.

Can a 'sales manager' of a recognised organisation be a professional referee?

I have never worked there but I know this person very well and vice versa since a very long time
